abstract

Cytochromes c of Acidithiobacillus ferrooxidans.

The chemolithoautotrophic Gram-negative bacterium Acidithiobacillus ferrooxidans is versatile and can grow on a number of electron donors and acceptors. In the A. ferrooxidans ATCC 23270 genome, computer analysis identified 11 genes encoding putative cytochromes c. At least eight putative cytochromes c were differentiated on gels in ATCC 33020 cells grown on ferrous iron or sulfur. All these cy...

Periplasmic Proteins of the Extremophile Acidithiobacillus ferrooxidans

Acidithiobacillus ferrooxidans is a chemolithoautotrophic acidophile capable of obtaining energy by oxidizing ferrous iron or sulfur compounds such as metal sulfides. Some of the proteins involved in these oxidations have been described as forming part of the periplasm of this extremophile. The detailed study of the periplasmic components constitutes an important area to understand the physiolo...

Phylogenetic heterogeneity of the species Acidithiobacillus ferrooxidans.

Polyphasic genotypic analysis of 25 Acidithiobacillus ferrooxidans strains isolated from ores and ore concentrates collected in different regions of the world showed considerable strain heterogeneity. Restriction patterns of the chromosomal DNA of these strains obtained by PFGE were specific for each strain. According to the degree of DNA relatedness, 17 of the 23 strains studied were divided i...

Organoarsenic resistance and bioremoval of Acidithiobacillus ferrooxidans.

The tolerance and bioremoval of dimethylarsinic acid (DMA(V)) by Acidithiobacillus ferrooxidans (A. ferrooxidans) were investigated here. The inhibitory concentration (IC) of DMA(V) was determined for A. ferrooxidans. The effects of various parameters such as pH, contact time, initial DMA(V) concentration, biosorbent dose and temperature were systematically examined to study the biosorption pro...

Reconstitution of iron oxidase from sulfur-grown Acidithiobacillus ferrooxidans.

The iron oxidation system from sulfur-grown Acidithiobacillus ferrooxidans ATCC 23270 cells was reconstituted in vitro. Purified rusticyanin, cytochrome c, and aa(3)-type cytochrome oxidase were essential for reconstitution. The iron-oxidizing activity of the reconstituted system was 3.3-fold higher than that of the cell extract from which these components were purified.

Thickness and surface density of extracellular polymers on Acidithiobacillus ferrooxidans.

In vivo force microscopy measurements of Acidithiobacillus ferrooxidans revealed a repulsive force that was due to the presence of extracellular polymers on the bacterium's surface. Measured force-distance profiles were fit to steric force theory to estimate the density and thickness values of these exopolymers. The polymer densities were 3.4 x 10(16) to 7.1 x 10(16) molecules m(-2), and the eq...
